Associations between farming practices and disease mortality of Pacific oyster Crassostrea gigas in a Mediterranean lagoon ArchiMer

We present the first large-scale, high-resolution spatial and temporal pattern of disease mortality caused by Ostreid herpesvirus 1 (OsHV-1) that has affected Pacific oyster Crassostrea gigas in France every year since 2008, in relation to local farm use. Mortality was monitored in healthy oyster spat deployed at 106 locations in the Thau Mediterranean lagoon during the epizootic that occurred in spring 2011. We conducted a field survey of farming practices in the areas where the sentinel oysters were placed before the epizootic. Created in 1992, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Ils activities are in keeping with two European Directives, 91167/EEC and 95170/EC and are a part of the institutional tasks of Ifremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to sm'vey the evolution of notifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (Granville bed and zone X). Depuis 1992, la surveillance de la santé des mollusques marins du littoral français est assurée par le réseau de Pathologie des Mollusques (Repamo). Ses activités s’inscrivent dans le cadre de la Directive Européenne 2006/88/CE. Depuis son évaluation par la plateforme nationale d’épidémiosurveillance en santé animale en 2012, l’objectif de surveillance est la détection précoce des infections dues à des organismes pathogènes exotiques et émergents affectant les mollusques marins sauvages et d’élevage. Oysters modify the planktonic microbial community structure by their filtration and NH4 excretion activities. While many studies have been conducted on this subject with adult oysters, none had been carried out in situ with juveniles. Pacific oyster juveniles (Magallana gigas, previously Crassostrea gigas) died massively all over the world since 2008 in relation with OsHV-1 infection. Since spat mortality events that were firstly detected in 2008, Mediterranean shellfish industry seeks to exploit the Pacific oyster spat from lagoon natural recruitment. The CRCM, Cepralmar and Ifremer developed accordingly a research project named PRONAMED Phase 1 and 2 - 2010-2014, in order to define the economic potential of spatfall and improve knowledge of the reproductive cycle of Crassostrea gigas in this specific environmental context. Mortality of oysters Crassostrea gigas occurred along the coasts of France during summer 2008, causing a serious crisis in French oyster production. During spring to summer 2008, 5 groups of 1-yr old oysters of different origins and ploidy levels deployed in the Thau lagoon (France) were sampled for biometrical and biochemical analyses; environmental parameters were monitored in the same area. Mortality occurred in 2 episodes: 13 May to 10 June and 24 June to 9 July. Wild-collected oysters showed mortality sooner than other groups but total overall mortality was 85% in all groups. Cette étude avait pour objectif principal de déterminer les facteurs explicatifs de la dynamique spatio-temporelle des surmortalités d’huîtres creuses dans la lagune de Thau. Les facteurs étudiés étaient : la pathologie (détection d’ADN appartenant à l’Ostreid herpesvirus 1 OsHV-1 et au groupe bactérien Vibrio splendidus), l’état énergétique des huîtres (triglycérides, sucres totaux et acides gras comme marqueurs trophiques), les pratiques d’élevage et l’hydrodynamique. Influence of OSHV-1 oyster mortality episode on dissolved inorganic fluxes: An ex situ experiment at the individual scale ArchiMer

Ostreid herpesvirus 1 (OsHV-1 μvar) infection has caused significant mortalities in juvenile oysters (Crassostrea gigas). In contrast to the practices of other animal production industries, sick and dead oysters are not separated from live ones and are left to decay in the surrounding environment, with unknown consequences on fluxes of dissolved materials. A laboratory approach was used in this study to test the influence of oyster mortality episode on dissolved inorganic fluxes at the oyster interface, dissociating (i) the effect of viral infection on metabolism of juvenile oysters and (ii) the effect of flesh decomposition on oxygen consumption and nutrient releases at the individual scale. We bring new items in connection with the origins of the natural recruitment variability of the Pacific oyster Crassostrea gigas in Thau lagoon. A poor relationship between " number of larvae" and "number of spat" in 2012 shows a biological lock around oysterculture areas during the metamorphosis in spite of better success of recruitment outside shellfish farming zone. A comprehensive analysis of environmental factors effects and the development of prognosis tools are planned to assist oysterfarmers in rationalization of natural oyster recruitment around Mediterranean lagoon.

The Thau Lagoon, from time to time is the area of crisis locally called « malaïgues ». The characteristics of these « malaïgues » are the lack of oxygen interesting all the water column and the white color of the waters during the phenomena. The result of this anoxy is the death of cultivated molluscs. From 1969 to 2000 nine « malaïgues » have been observed. They occur during the Summer period but in different locations. The surface of the areas concerned are very different according the year. Different means of struggle have been tried and are described.
